Você or tu?

Você, almost everywhere in Brazil. Tu survives in the south and the northeast but takes different verb endings, which is why it is not in this set. O senhor and a senhora are the formal step up, for someone much older or in a position of authority.

Why is there a tilde on não?

It nasalises the vowel, and it is not optional. Portuguese uses the tilde far more than Spanish does, and the same mark turns pao into pão and mae into mãe. Leaving it off changes the word rather than just the spelling.
